Poly Acrylamide Gel Electrophoresis method
How do the different plasmid forms effect their mobility through gel electrophoresis?
The plasmid in your preparation exists in a number of different forms (supercoiled, catanane, open-circular). Which form do you think will migrate the fastest? (I think it is supercoiled, based on the fact that smaller, more tightly-packed DNA travels faster through the gel matrix). Is your intuition correct? (Yes). To answer this question will require some research into the theory and mathematics underlying electrophoretic procedures. (Huh? This is where I am currently stuck, as I really cannot find a link that can provide the specific/general kind of information I need). Please help! :s
Charge and size is all you've got in electrophoresis. Charge is fixed for a given bit of DNA, so all your decision depends on is size (i.e., viscous drag through the gel).
